RGD targeting of human ferritin iron-oxide nanoparticles enhances in vivo molecular MRI of experimental aortic aneurysms

چکیده
Background Both inflammation and angiogenesis contribute to the progression of abdominal aortic aneurysm (AAA) disease. RGD is a peptide binder of the avb3 integrin, which is expressed highly on activated macrophages and angiogenic endothelial cells. Human ferritin (HFn) is a nanoscale protein cage with 12nm diameter and 8nm interior cavity, which we have utilized as a platform for molecular/cellular imaging. We can genetically introduce RGD peptide to HFn. The purpose of this study is to evaluate RGD-conjugated HFn iron oxide nanoparticles for enhanced in vivo MRI of murine AAAs.

PURPOSE To evaluate Arg-Gly-Asp (RGD)-conjugated human ferritin (HFn) iron oxide nanoparticles for in vivo magnetic resonance imaging (MRI) of vascular inflammation and angiogenesis in experimental carotid disease and abdominal aortic aneurysm (AAA). MATERIALS AND METHODS HFn was genetically engineered to express the RGD peptide and Fe3 O4 nanoparticles were chemically synthesized inside the ...

2) RGD-conjugated HFn-iron oxide nanoparticles Using the recombinant human heavy-chain ferritin protein cage, HFn was genetically engineered to display 24 copies of an RGD-4C peptide (CDCRGDCFC) on the exterior surface of the protein cage. Magnetite (Fe3O4) was encapsulated in interior cavities of RGD-conjugated HFn (RGD HFn) and non-targeted HFn (RGD HFn) at loading factors of 5000Fe per cage,...

PURPOSE Arginine-glycine-aspartic acid (RGD)-based nanoprobes allow specific imaging of integrin αvβ3, a protein overexpressed during angiogenesis. Therefore, this study applied a novel RGD-coupled, polyacrylic acid (PAA)-coated ultrasmall superparamagnetic iron oxide (USPIO) (referred to as RGD-PAA-USPIO) in order to detect tumor angiogenesis and assess the early response to antiangiogenic tre...

UNLABELLED The purpose of this study was to develop a bifunctional iron oxide (IO) nanoparticle probe for PET and MRI scans of tumor integrin alphavbeta3 expression. METHODS Polyaspartic acid (PASP)-coated IO (PASP-IO) nanoparticles were synthesized using a coprecipitation method, and particle size and magnetic properties were measured. A phantom study was used to assess the efficacy of PASP-...
